NM_000527.5(LDLR):c.368C>G (p.Ser123Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S123C variant (also known as c.368C>G), located in coding exon 4 of the LDLR gene, results from a C to G substitution at nucleotide position 368. The serine at codon 123 is replaced by cysteine, an amino acid with dissimilar properties. This variant was reported in a hypercholesterolemia cohort (Tada H et al. J Clin Lipidol, 2022 Sep;16:863-869). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
