Uncertain significance for EGFR-related lung cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005228.5(EGFR):c.14G>A (p.Gly5Glu), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change replaces glycine with glutamic acid at codon 5 of the EGFR protein (p.Gly5Glu). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and glutamic acid. This variant has not been reported in the literature in individuals affected with EGFR-related conditions.
