Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005869.4(CWC27):c.1151A>G (p.Gln384Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with arginine at codon 384 of the CWC27 protein (p.Gln384Arg). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with CWC27-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
