NM_177438.3(DICER1):c.2108A>G (p.His703Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H703R variant (also known as c.2108A>G), located in coding exon 12 of the DICER1 gene, results from an A to G substitution at nucleotide position 2108. The histidine at codon 703 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
