Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_017617.5(NOTCH1):c.6257A>G (p.Asn2086Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: NOTCH1 c.6257A>G (p.Asn2086Ser) results in a conservative amino acid change located in the Ankyrin repeat region (IPR002110) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.1e-06 in 1605292 control chromosomes (gnomAD v4.1). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.6257A>G in individuals affected with Adams-Oliver Syndrome 5 and no experimental evidence demonstrating its impact on protein function have been reported.
