This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 737 through coding-DNA position 739, duplicating 3 bases; at the protein level this means converts the codon for serine at residue 247 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ser247*) in the IFIH1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in IFIH1 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
