Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.1634A>C (p.Glu545Ala), citing Ambry Variant Classification Scheme 2023: The p.E545A variant (also known as c.1634A>C), located in coding exon 15 of the POLE gene, results from an A to C substitution at nucleotide position 1634. The glutamic acid at codon 545 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
