Uncertain significance for Microcephaly, normal intelligence and immunodeficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002485.5(NBN):c.1990A>T (p.Ile664Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with phenylalanine at codon 664 of the NBN protein (p.Ile664Phe). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NBN-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
