Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139057.4(ADAMTS17):c.2225G>A (p.Arg742His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with histidine at codon 742 of the ADAMTS17 protein (p.Arg742His). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (no rsID available, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with ADAMTS17-related conditions. ClinVar contains an entry for this variant (Variation ID: 1498607). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
