This variant lies in the MTM1 gene (transcript NM_000252.3) at coding-DNA position 913, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 305 with glutamine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 305 of the MTM1 protein (p.Glu305Gln). This variant is present in population databases (rs587783861, gnomAD 0.004%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with MTM1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1498542).
