Uncertain significance for Immunodeficiency 35 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003331.5(TYK2):c.3520G>A (p.Glu1174Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 1174 of the TYK2 protein (p.Glu1174Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (no rsID available, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TYK2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1497952). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
