Uncertain significance for Primary immunodeficiency with post-measles-mumps-rubella vaccine viral infection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005419.4(STAT2):c.1418A>C (p.Asn473Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with threonine at codon 473 of the STAT2 protein (p.Asn473Thr). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and threonine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt STAT2 protein function. This variant has not been reported in the literature in individuals with STAT2-related conditions. This variant is not present in population databases (ExAC no frequency).
