NM_007294.4(BRCA1):c.4153C>G (p.Leu1385Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1385V variant (also known as c.4153C>G), located in coding exon 10 of the BRCA1 gene, results from a C to G substitution at nucleotide position 4153. The leucine at codon 1385 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
