NM_017617.5(NOTCH1):c.6119A>G (p.Asn2040Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N2040S variant (also known as c.6119A>G), located in coding exon 33 of the NOTCH1 gene, results from an A to G substitution at nucleotide position 6119. The asparagine at codon 2040 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
