This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 4533, where G is replaced by C; at the protein level this means replaces glutamine at residue 1511 with histidine — a missense variant. Submitter rationale: This sequence change replaces glutamine with histidine at codon 1511 of the DSP protein (p.Gln1511His). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DSP-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
