Likely pathogenic for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005359.6(SMAD4):c.1598T>C (p.Leu533Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 533 of the SMAD4 protein (p.Leu533Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of juvenile polyposis syndrome and hereditary hemorrhagic telangiectasia (PMID: 20101697; internal data). ClinVar contains an entry for this variant (Variation ID: 1495356). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
