Likely pathogenic for Sandhoff disease — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000521.4(HEXB):c.1508+1G>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the HEXB gene (transcript NM_000521.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1508, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: HEXB c.1508+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: three predict the variant weakens a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 251356 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1508+1G>A in individuals affected with Sandhoff Disease and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as likely pathogenic. Based on the evidence outlined above, the variant was classified as likely pathogenic.
