Uncertain significance for 2-aminoadipic 2-oxoadipic aciduria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018706.7(DHTKD1):c.188A>T (p.Tyr63Phe), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs768895085, ExAC 0.001%). This sequence change replaces tyrosine with phenylalanine at codon 63 of the DHTKD1 protein (p.Tyr63Phe). The tyrosine residue is highly conserved and there is a small physicochemical difference between tyrosine and phenylalanine. This variant has not been reported in the literature in individuals affected with DHTKD1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The phenylalanine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
