Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000782.5(CYP24A1):c.544-17G>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 3 of the CYP24A1 gene. It does not directly change the encoded amino acid sequence of the CYP24A1 protein. This variant is present in population databases (rs373305362, gnomAD 0.01%). This variant has been observed in individual(s) with clinical features of CYP24A1-related conditions (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1494971). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
