NM_001378454.1(ALMS1):c.6612G>C (p.Arg2204Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R2205S variant (also known as c.6615G>C), located in coding exon 8 of the ALMS1 gene, results from a G to C substitution at nucleotide position 6615. The arginine at codon 2205 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
