Uncertain significance for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001849.4(COL6A2):c.2797C>T (p.Arg933Cys), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with cysteine at codon 933 of the COL6A2 protein (p.Arg933Cys). The arginine residue is weakly conserved and there is a large physicochemical difference between arginine and cysteine. This variant has not been reported in the literature in individuals with COL6A2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
