Uncertain significance for Inflammatory bowel disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000572.3(IL10):c.121C>T (p.Leu41Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with phenylalanine at codon 41 of the IL10 protein (p.Leu41Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant is present in population databases (rs750010814, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with IL10-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
