This variant lies in the SYNE1 gene (transcript NM_182961.4) at coding-DNA position 6646, where T is replaced by C; at the protein level this means replaces cysteine at residue 2216 with arginine — a missense variant. Submitter rationale: This sequence change replaces cysteine with arginine at codon 2223 of the SYNE1 protein (p.Cys2223Arg). The cysteine residue is weakly conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SYNE1-related conditions.
