This variant lies in the WDR45 gene (transcript NM_001029896.2) at coding-DNA position 482, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 161 with serine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine with serine at codon 162 of the WDR45 protein (p.Phe162Ser). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with WDR45-related conditions. This missense change has been observed in at least one individual who was not affected with WDR45-related conditions (Invitae).
