Uncertain significance for Autoimmune lymphoproliferative syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000639.3(FASLG):c.313T>C (p.Phe105Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with leucine at codon 105 of the FASLG protein (p.Phe105Leu). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with FASLG-related conditions. This variant is not present in population databases (ExAC no frequency).
