Uncertain significance for Familial episodic pain syndrome with predominantly lower limb involvement; Hereditary sensory and autonomic neuropathy type 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001349253.2(SCN11A):c.4113del (p.Ile1372fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ile1372Serfs*2) in the SCN11A gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SCN11A cause disease. This variant is present in population databases (no rsID available, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with SCN11A-related conditions. ClinVar contains an entry for this variant (Variation ID: 1493523). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
