Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_138413.4(HOGA1):c.290G>T (p.Arg97Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HOGA1 c.290G>T (p.Arg97Leu) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4e-06 in 251454 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.290G>T in individuals affected with HOGA1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. Two other variants at this residue have been reported as Pathogenic/Likely Pathogenic (c.290G>A, p.Arg97His and c.289C>T, p.Arg97Cys), suggesting that this codon is functionally important.
