Uncertain significance for Familial focal epilepsy with variable foci — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001242896.3(DEPDC5):c.4715T>A (p.Leu1572Gln), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with DEPDC5-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with glutamine at codon 1572 of the DEPDC5 protein (p.Leu1572Gln). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and glutamine. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C65"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
