Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.176C>T (p.Ser59Phe), citing Ambry Variant Classification Scheme 2023: The p.S59F variant (also known as c.176C>T), located in coding exon 2 of the BRIP1 gene, results from a C to T substitution at nucleotide position 176. The serine at codon 59 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
