This variant lies in the PRKDC gene (transcript NM_006904.7) at the canonical splice acceptor site of the intron immediately before coding-DNA position 7455, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 55 of the PRKDC gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in PRKDC cause disease.
