This variant lies in the MCCC1 gene (transcript NM_020166.5) at the canonical splice donor site of the intron immediately after coding-DNA position 1681, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The MCCC1 c.1681+2T>C variant is predicted to disrupt the GT donor site and interfere with normal splicing. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0065% of alleles in individuals of South Asian descent in gnomAD (http://gnomad.broadinstitute.org/variant/3-182751777-A-G). Variants that disrupt consensus splice donor sites in MCCC1 are expected to be pathogenic. Therefore, we interpret this variant as pathogenic.
