This variant lies in the TUBA1A gene (transcript NM_006009.4) at coding-DNA position 53, where A is replaced by T; at the protein level this means replaces asparagine at residue 18 with isoleucine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces asparagine with isoleucine at codon 18 of the TUBA1A protein (p.Asn18Ile). The asparagine residue is highly conserved and there is a large physicochemical difference between asparagine and isoleucine. This variant has been observed in individual(s) with clinical features of TUBA1A-related cortical malformations (Invitae). In at least one individual the variant was observed to be de novo. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
