Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020999.4(NEUROG3):c.53A>T (p.Glu18Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 18 of the NEUROG3 protein (p.Glu18Val). This variant is present in population databases (rs747663752, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with NEUROG3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
