Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006506.5(RASA2):c.2365C>T (p.Gln789Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gln789*) in the RASA2 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in RASA2 cause disease. This variant is present in population databases (rs760281563, ExAC 0.002%). This variant has not been reported in the literature in individuals with RASA2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
