Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014314.4(RIGI):c.149C>A (p.Pro50Gln), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with DDX58-related conditions. This variant is present in population databases (rs533959324, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This sequence change replaces proline, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 50 of the DDX58 protein (p.Pro50Gln). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. ClinVar contains an entry for this variant (Variation ID: 1490091).
