Uncertain significance for Short-rib thoracic dysplasia 14 with polydactyly; Joubert syndrome 23 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001329943.3(KIAA0586):c.411-1369A>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the KIAA0586 gene (transcript NM_001329943.3) at 1369 bases into the intron immediately before coding-DNA position 411, where A is replaced by G. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 188 of the KIAA0586 protein (p.Asp188Gly). This variant is present in population databases (no rsID available, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with KIAA0586-related conditions. ClinVar contains an entry for this variant (Variation ID: 1489760). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
