NM_000263.4(NAGLU):c.1880C>T (p.Ala627Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1880C>T (p.A627V) alteration is located in exon 6 (coding exon 6) of the NAGLU gene. This alteration results from a C to T substitution at nucleotide position 1880, causing the alanine (A) at amino acid position 627 to be replaced by a valine (V). Based on data from gnomAD, the T allele has an overall frequency of 0.008% (22/261220) total alleles studied. The highest observed frequency was 0.092% (21/22742) of African alleles. This amino acid position is not well conserved in available vertebrate species. In an assay testing NAGLU function, this variant showed a functionally abnormal result (Clark, 2018). This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
