Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003816.3(ADAM9):c.1892A>G (p.Asn631Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with serine at codon 631 of the ADAM9 protein (p.Asn631Ser). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and serine. This variant is present in population databases (rs532248416, ExAC 0.05%). This variant has not been reported in the literature in individuals affected with ADAM9-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
