Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007348.4(ATF6):c.1618A>G (p.Ser540Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 540 of the ATF6 protein (p.Ser540Gly). This variant is present in population databases (rs761144728, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with ATF6-related conditions. ClinVar contains an entry for this variant (Variation ID: 1489226). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
