Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001039141.3(TRIOBP):c.2063G>A (p.Ser688Asn), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The asparagine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This variant has not been reported in the literature in individuals affected with TRIOBP-related conditions. This variant is present in population databases (rs748003804, ExAC 0.009%). This sequence change replaces serine with asparagine at codon 688 of the TRIOBP protein (p.Ser688Asn). The serine residue is weakly conserved and there is a small physicochemical difference between serine and asparagine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
