Uncertain significance for Surfactant metabolism dysfunction, pulmonary, 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172245.4(CSF2RA):c.461T>C (p.Ile154Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with threonine at codon 154 of the CSF2RA protein (p.Ile154Thr). The isoleucine residue is moderately conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with CSF2RA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
