Uncertain significance for Familial melanoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_058195.4(CDKN2A):c.47G>A (p.Gly16Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with aspartic acid at codon 16 of the CDKN2A (p14ARF) protein (p.Gly16Asp). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with cutaneous malignant melanoma (PMID: 15937071). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Experimental studies have shown that this missense change does not substantially affect CDKN2A (p14ARF) protein function (PMID: 15937071).
