NM_000179.3(MSH6):c.64A>C (p.Lys22Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K22Q variant (also known as c.64A>C), located in coding exon 1 of the MSH6 gene, results from an A to C substitution at nucleotide position 64. The lysine at codon 22 is replaced by glutamine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
