Uncertain significance for Granulomatous disease, chronic, autosomal recessive, cytochrome b-positive, type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000433.4(NCF2):c.613G>A (p.Val205Met), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NCF2 gene (transcript NM_000433.4) at coding-DNA position 613, where G is replaced by A; at the protein level this means replaces valine at residue 205 with methionine — a missense variant. Submitter rationale: This sequence change replaces valine with methionine at codon 205 of the NCF2 protein (p.Val205Met). The valine residue is highly conserved and there is a small physicochemical difference between valine and methionine. This variant is present in population databases (rs761985515, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with NCF2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
