This variant lies in the SYNJ1 gene (transcript NM_203446.3) at 152 bases past the stop codon (3' untranslated region), where T is replaced by C. Submitter rationale: This sequence change replaces leucine with serine at codon 1394 of the SYNJ1 protein (p.Leu1394Ser). The leucine residue is moderately conserved and there is a large physicochemical difference between leucine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SYNJ1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
