Uncertain significance for Leber congenital amaurosis 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014336.5(AIPL1):c.983G>C (p.Ser328Thr), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with threonine at codon 328 of the AIPL1 protein (p.Ser328Thr). The serine residue is weakly conserved and there is a small physicochemical difference between serine and threonine. This variant has not been reported in the literature in individuals affected with AIPL1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
