Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020884.7(MYH7B):c.235A>G (p.Asn79Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with aspartic acid at codon 121 of the MYH7B protein (p.Asn121Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid. This variant is present in population databases (rs774618536, ExAC 0.009%). This variant has not been reported in the literature in individuals with MYH7B-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
