NM_001289808.2(CRYAB):c.148C>T (p.Arg50Trp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R50W variant (also known as c.148C>T), located in coding exon 1 of the CRYAB gene, results from a C to T substitution at nucleotide position 148. The arginine at codon 50 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
