Uncertain significance for Hypertrophic cardiomyopathy 12; Dilated cardiomyopathy 1M — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003476.5(CSRP3):c.468del (p.Thr157fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 468, deleting one base; at the protein level this means shifts the reading frame starting at threonine residue 157, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a frameshift in the CSRP3 gene (p.Thr157Glnfs*51). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 38 amino acid(s) of the CSRP3 protein and extend the protein by 12 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CSRP3-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
