NM_152564.5(VPS13B):c.3249T>A (p.Asp1083Glu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1083E variant (also known as c.3249T>A), located in coding exon 22 of the VPS13B gene, results from a T to A substitution at nucleotide position 3249. The aspartic acid at codon 1083 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
